A Youtube site, e.g. an exact YouTube clone, is not that fundamentally hard to design.

At its most basic level:
Its comprised of a user registration system.
An upload component that allows users to upload videos
SQL Server indexing of the videos (tags, keywords, etc)
Pages that can dynamically embed a video.

For that matter, it's probably not much different fundamentally than the Wrox Photo Album example in chapter 7 of ASP.NET Instant Results:

Whether your users are uploading photos or videos, ASP.NET really doesn't care. Same difference.

how to design

because i never do this type of job

Dealing with video is not really an issue of ASP.NET, it's a matter of knowing the right HTML syntax to provide the browser with the right information to handle the resource you wish it to handle (whether it's a img, link to a page, video, etc). Try this to start: http://www.google.com/search?q=embed+tag+video
