Hmmm, in that case, it's strange.
For the update to work successfully, the account that IIS is running under needs to write some stuff to the Database folder. So, that's why you need to set the permissions to the folder, rather than to the database. So, in your case, that seems fine.
Two other things I can think of:
1. Did you close Access?? If you still have Access with your database open in a design window, you'll get this error as well.
2. Are you sure IIS is using the IUSR_MachineName account? You can do a quick check, by assigning the MachineName\Users or Everyone group full access to the database folder. If the code then works, it's a permission issue. Don't forget to remove the Users group right away, as you don't want everyone to mess with your databases.
