Yes indeed it does! Perhaps an hour or more!
I ran a number of commands from the app dev vm as well as the server.
Also looking at the technet entry for this cmdlet

I also ran a focused replication on just my app dev vm since the Lync trial vm set since the topology entry in the Lync control panel includes a server (qms) that I had turned off and is listed as failed replication. It probably wouldn't block my app dev server although since the server that is turned off is first on the list, and the command took so long I wanted to be sure that the content store replication command would succeed.
