error 404 could not open a connection to sql serve

Hi,
I tried the
"Data source=.\SQLEXPRESS;AttachDbFilename=C:\Document and settings\user1\My Documents\User.mdf;Integrated Security=True;Connect Timeout=30;User Instance=True"
in a web application using c#,net2005.
Can u help me to sortout this problem.
