How to generate 12 random questions

could someone help me how to create and generate 12 random questions when we are creating code in vb.net i mean file handling.

Perhaps you can clarify your question a bit?

What is it you are trying to do?
Is this a little test application, where the user will see some questions that need to answer?
Do you have a file that contains a number of questions from which you want to select 12 questions at random to display to a use - perhaps one at a time?

Please clarify.

this is a test application where the users will need to answer 12 questions random.

Well... a full source code example for that might be something you can find by doing a simple search of the internet.
